Bioavailability challenges. Semaglutide is a peptide. Peptides are normally destroyed by stomach acid and digestive enzymes. Rybelsus solves this with a co-formulated absorption enhancer (sodium N-(8-(2-hydroxybenzoyl)amino)caprylate, or SNAC), which temporarily neutralizes stomach acid and allows some semaglutide to be absorbed intact. Even so, oral bioavailability is roughly 0.4-1% of the dose, requiring much larger oral doses than injection.
Novo Nordisk ran the OASIS phase 3 trial program testing higher-dose oral semaglutide (25 mg and 50 mg daily) for weight loss in adults without diabetes. Results published in 2023 showed meaningful weight reduction, but as of 2026 FDA approval for a higher-dose oral weight-loss formulation has not occurred. If approved, it would likely be marketed under a new brand name rather than as “oral Wegovy.”

How Wegovy Injections Work
Wegovy is sold in a single-dose pen with a fixed dose preloaded. You do not need to draw up the medication. The injection is given subcutaneously (just under the skin) in the abdomen, thigh, or upper arm, once per week. The needle is short and most people report minimal pain.
Dosing follows a slow titration schedule to minimize gastrointestinal side effects:
